Research activity
Our research activity is been mainly focused on the role of advanced glycation end products (AGEs), oxidative stress and inflammation in the development of chronic diseases. AGEs are reactive metabolites deriving from exogenous sources or endogenous reactions between reducing sugars or fats and proteins. The tissue accumulation of AGEs is a hallmark of physiological ageing and is correlated to the gradual development of a low grade chronic inflammation, the so called “inflammaging” taking part in most of the age-related diseases. Recently we demonstrated that adult survivors of pediatric cancer which have been exposed to chemotherapy and radiotherapy in the youth display higher levels of plasma AGEs and AGEreceptors, as well as an altered plasma inflammatory and lipid profile that may be involved in the increased risk for early cardiovascular events extensively reported in cancer survivors.
We previously demonstrated in different models in animals models of diet-induced metabolic diseases that AGEs are critical drivers of alterations in many aspects of glucose and lipid metabolism, contributing, through direct and indirect mechanisms, to a general pro-inflammatory condition and energy metabolism impairment thanks to inter-organs crosstalk involving the gut, the liver, the kidney and the brain, thus suggesting their potential role in age-related morbidities, ranging from cardiovascular to neurodegenerative diseases.Another project in the starting phase thanks to a funding from the CRT 2022 Foundation
- HOdgkinLymphoma Disease survivors STUDY: AGE, inflammation and aging which received
a positive opinion from the ethics committee (00096/2023), relating to the evaluation
of oxidative stress, inflammaging and accumulation of AGEs and correlate them with the
presence of clinical and biochemical markers of premature aging, in a cohort of cancer
survivors treated for pediatric or young adult Hodgkin's lymphoma and compare them with
a control population consisting of age-matched subjects and one consisting of
adult-elderly subjects. Hodgkin's lymphoma was chosen for its number within the CAYACS
cohort currently in follow-up of the structures participating in the project, for the
high cumulative incidence of LE that characterizes these survivors and for the relative
homogeneity of the anticancer treatments used . Recently, the group has activated a collaboration with Prof. Ciocan
(Occupational medicine) in the evaluation of biological indicators of aging and
analysis of occupational parameters in a cohort of workers belonging to the
occupational medicine clinic with the relative approval of the Ethics Committee.

The main object of our research is to evaluate molecular pathways affected by AGEs that are responsible for the onset of premature age-related diseases (i.e. epigenetic modifications of genes involved in pro-inflammatory/defensive pathways; mitochondrial dysfunction)
ATTIVITÀ DI RICERCA
La nostra attività di ricerca si è concentrata principalmente sul ruolo dei prodotti terminali della glicazione avanzata (AGE), dello stress ossidativo e dell'infiammazione nello sviluppo di malattie croniche. Gli AGE sono metaboliti reattivi derivanti da fonti esogene o reazioni endogene tra zuccheri riducenti o grassi e proteine. L'accumulo tissutale di AGE è un segno distintivo dell'invecchiamento fisiologico ed è correlato allo sviluppo graduale di un'infiammazione cronica di basso grado, la cosiddetta “meta infiammazione” che è presente nella maggior parte delle malattie legate all'età.
Recentemente abbiamo dimostrato che gli adulti sopravvissuti al linfoma di Hodgkin in età pediatrica che sono stati esposti a chemioterapia e radioterapia mostrano livelli più elevati di AGE plasmatici, nonché un profilo infiammatorio e lipidico plasmatico alterato che pu essere correlato all’'aumentato rischio cardiovascolare precoce ampiamente riportato nei sopravvissuti al linfoma. Abbiamo precedentemente dimostrato, in diversi modelli animali di malattie metaboliche indotte da dieta che gli AGE sono determinanti critici delle alterazioni del metabolismo del glucosio e dei lipidi, contribuendo, attraverso meccanismi diretti e indiretti, ad una condizione pro-infiammatoria sistemica e al deterioramento del metabolismo energetico grazie al cross-talk tra i vari organi, suggerendo così un loro potenziale ruolo nelle differenti morbilità legate all'età, che vanno dalle malattie cardiovascolari a quelle neurodegenerative. Altro progetto in fase di partenza grazie ad un finanziamento della Fondazione CRT 2022- HOdgkinLymphoma Disease survivors STUDY: AGE, inflammation and aging che ha avuto parere positivo dal comitato etico (00096/2023), relativo alla valutazione di stress ossidativo, inflammaging e l’accumulo di AGE e correlarli con la presenza di marcatori clinici e biochimici di aging prematuro, in una coorte di cancer survivors curati per linfoma di Hodgkin in età pediatrica o giovane adulta e confrontarli con una popolazione di controllo costituita da soggetti pari età ed una costituita da soggetti in età adulto-anziana. Il linfoma di Hodgkin è stato scelto per la sua numerosità all’interno della coorte di CAYACS attualmente in follow-up le strutture partecipanti al progetto, per l’elevata incidenza cumulativa di LE che caratterizza questi survivors e per la relativa omogeneità dei trattamenti antitumorali impiegati.
Recentemente il gruppo ha attivato una collaborazione con la Prof.ssa Ciocan (Medicina del lavoro) nella valutazione degli indicatori biologici di invecchiamento e analisi dei parametri lavorativi in una coorte di lavoratori afferenti all’ambulatorio di Medicina del Lavoro con relativa approvazione del Comitato etico. Partecipanti allo studio: eleonora Aimeretti e la tesista Atefeh Moradi.
L'obiettivo in tutti gli studi rimane quello di valutare il ruole chiave degli AGEs responsabili di scatenere l'insorgenza di prematura mallattie correlate con l'invecchiamento
